| The PPAR alpha, important regulator of fatty acid metabolism in heart shown to be involved in development of cardiovascular disease. The radiation induced impairment of fatty acid metabolism was observed due to increased phosphorylation of PPAR alpha that leads to reduction in the expression of key proteins involved in fatty acid oxidation and transport with 16 Gy irradiation after 16 weeks and 40 weeks in C57BL/6J mice. The aim of our present study is to examine the influence of PPAR-α in the development of different cardiovascular disease phenotype after high dose local ionizing radiation in heart after long term exposure compared to wild type mice and the role of PPAR-α in this disease process. Male mice strain (C57BL/6J) with different genotype for PPAR-α; PPAR-α wild type, PPAR-α +/- (Heterozygous mutant), PPAR-α -/- (Homozygous mutant), were irradiated locally on the heart with single X-ray doses of 8 and 16 Gy at the age of 8 weeks. Radiation induced effects on heart from different PPAR-α genotypes were analyzed by quantitative proteomics after 20 weeks of exposure. Further proteomics data were validated using bioinformatics tools, western blotting, immunohistochemistry, election microscopy, and ELISA. | ||
